643 Immunoprevention of dysplastic nevi in mice

Melanoma, an aggressive malignancy of melanocytes, is responsible for more deaths than any other skin cancer. Despite the fact that there a long lag period before premalignant dysplastic nevi become invasive melanomas, has been little progress in developing new methods melanoma prevention. Using animal model melanomagenesis, we examined whether administration anti-PD-L1 antibodies, effective immunotherapy advanced and metastatic melanoma, could be employed immunoprevention. When mice were pretreated with Abs to PD-L1 prior topical regimen produces (treatment C3H/HeN DMBA TPA), significantly fewer developed at 25 weeks compared animals given isotype control Abs. Treatment ant-PD-L1 was associated significant increase CD8+ T-cells produced IL-17 (0.4% vs. 0.78%) IFN-γ (0.98 2.47%) had controls. There also decrease CD11b+, Gr1+ (3.28% 1.84%) CD4+CD25+Foxp3+ (8.93 3.56) cells. These findings indicate treatment may immunoprevention individuals who are risk melanoma.
